HT-29 Cells: The Multitalented Workhorse of Colorectal Research
The HT-29 cell line was established in 1964 from a primary colorectal adenocarcinoma of a 44-year-old Caucasian woman. It is an adherent human colorectal adenocarcinoma cell line — and one of the oldest and most widely used cell lines in colorectal cancer research.
Yet HT-29 is far more than a cancer model. Its real value lies in its multiplex identity: under the right conditions, it can act as a tumor cell, a surrogate for intestinal absorptive cells, and even a stand-in for mucus-secreting goblet cells. This plasticity makes it a powerful platform for intestinal biology, drug transport, and host–microbe studies.

Intestinal Epithelial Cell Properties
HT-29 expresses many intestinal epithelial markers — including keratins — and can form functional tight junctions, closely mimicking the epithelial characteristics of the gut barrier.
Mucus Secretion
This is one of HT-29's most striking features. Under specific culture conditions (for example, treatment with metronidazole or dexamethasone), HT-29 cells differentiate into goblet-like cells that secrete large amounts of mucins. This makes the line an excellent model for studying the intestinal mucus layer and goblet cell function.
Brush Border Enzyme Expression
HT-29 can express and secrete multiple intestinal brush border enzymes, including alkaline phosphatase and sucrase-isomaltase. Expression of these enzymes increases markedly once the cells reach confluence, reflecting enterocyte-like maturation.
Multilineage Differentiation Potential
Beyond goblet cells, HT-29 can display characteristics of other intestinal epithelial cell types under different induction conditions — evidence of its utility as a de facto model of intestinal stem cell behavior.

In drug transport studies, the intact HT-29 monolayer is relatively more permeable — and therefore less "tight" — than the well-known Caco-2 model. But once differentiated, it offers unique insight into how the mucus layer influences drug absorption, complementing Caco-2 data from an entirely different angle.
It Has a Large "Family"
Like Jurkat cells, decades of serial passaging have introduced subtle genetic drift among HT-29 stocks in labs worldwide, giving rise to distinct sublines that may differ slightly in growth rate and differentiation capacity.
